Immune tolerance was thought to prevent self-directed immune reactions from becoming a major cause of disease.
What we know now
Antibodies are immune proteins that recognize specific targets. Those targets are often foreign, such as parts of microbes or their toxins, but antibodies and immune cells can also recognize the body’s own tissues. Normally the immune system restrains these reactions; when that control fails, autoimmune disease can result.
Why it changed
Early immunology was strongly influenced by Paul Ehrlich’s idea of horror autotoxicus: the expectation that organisms had safeguards against harmful immune attacks on themselves. Researchers later found antibodies that reacted with the body’s own tissues and showed experimentally that self-directed immune responses could cause disease.
